Fluorescent recovery in the lower chamber (well) was measured after 15 minutes as stated in Section 2. Solute permeability (b) was assessed by adding 100 l of 1 mg/mL fluoresceinated dextran-70 in Transwell TM inserts. The Millicell® ERS-2 system uses alternating current (AC) to make membrane resistance measurements. Organic electrochemical transistor (OECT), in particular, outperforms many biosensors because of its multiple advantages such as high transconductance. Due to the small size of the electrodes, the user can easily measure transepithelial voltage and the resistance of cells grown on microporous membranes. Hence, several scientific techniques, such as epithelial voltohmmeter (EVOM) and electric cell-substrate impedance sensing (ECIS), were developed to assess barrier function for in vitro assays. The unique electronic circuit of the EVOM2 and the included STX2 elec-trode detect the confluence of the cellular monolayer. The EVOM2 not only qualitatively measures cell monolayer health, but also quantitatively measures cellular confluence. A silver/silver chloride (Ag/AgCl) pellet on each electrode tip measures voltage. EVOM2 is the next generation, redesigned for ease of use. WPI experimented with two volt-ohm meters: An expensive Fluke meter applies a DC voltage of 50 to 80mV, which (in a 1000 membrane) will pass 80A. An increase in TEER detected with the electronic circuit of the Millicell® ERS-2 meter and its electrode is an indication of cell monolayer health and confluence. A volt-ohm meter may apply a constant voltage of unknown current across the membrane and damage the cells electrically and also leave a chemical imbalance in the electrodes. This device qualitatively measures cell monolayer health and quantitatively measures cell confluence. ![]() The Millicell® ERS (Electrical Resistance System) reliably measures membrane potential and resistance of epithelial cells in culture. ![]()
